Designed for individuals preparing genomic and/or amplicon DNA libraries for sequencing on the Ion Torrent™ Personal Genome Machine™ and seeking detailed knowledge of the molecular biology techniques used for DNA library construction. Attendees will benefit by having training in a class environment, hands-on lab practicals, and interaction with fellow attendees and Ion Torrent application scientists during lecture, discussion, and lab sessions.
Prerequisites:This class assumes attendees have prior knowledge of biological systems and basic experience with laboratory techniques such as pipetting and PCR.
Course Description:A 1-day course involving classroom and laboratory sections for DNA library construction for sequencing on the Ion Torrent™ Personal Genome Machine™. Attendees will gain hands-on experience in constructing a DNA fragment library using control genomic DNA, and learn about different methods for shearing and size-selection, library quality assessment, amplification, quantification and troubleshooting. Class discussion also reviews amplicon library construction. (Please note, we will NOT be able to use customer samples for library construction during this course.) Information and strategies for different sequencing applications and how these relate to library construction and preparation of genomic samples will also be discussed.

Ideally, individuals have already participated in the live, online System and Library Training, although this is not mandatory. If this is the first training after instrument purchase, it is mandatory to have your instrument fully operational; being able to use the instrument right after the training guarantees the best use of your time. This class assumes attendees have prior knowledge of biological systems and basic experience with laboratory techniques such as pipetting and PCR.

Course topics include an overview of instrument operation and hardware, library preparation, Ion Sphere™ particles preparation and deposition, quality control runs, troubleshooting, and data analysis. Hands-on training includes initiating and monitoring sequencing runs during class, and creating Ion Sphere™ particles for template preparation using the Ion OneTouch™ System. Course includes in depth discussions of RNA preparation, adapter ligation, reverse transcription, size selection, amplification, and quantification. Construction of SAGE™ libraries is not included in this course.

Course Description:This web session will provide an introduction to the Ion Reporter™ Software and show users how to analyze data acquired from Ion Torrent PGM™ or Proton™ Systems. Each session will include a 30 minute presentation and demo with a 20-30 minute open discussion.
